保山地区单采血小板献血员铁蛋白水平调查

目的 调查本地单采血小板献血员的血清铁蛋白水平,关爱献血员健康,提供更好的献血服务。方法 对保山地区近两年424例单采血小板献血员进行铁蛋白检测,按献血频次、性别、年龄分析血小板单采对铁蛋白的影响情况。结果 424例样本中铁蛋白低值率为:12.3%。其中男性:13.1%,女性:9.1%,二者差异无统计学意义(P>0.05),25~49岁年龄组和>50岁年龄组铁蛋白低值率高于18~24岁年龄组,且差异有统计学意义(P<0.05),单采血小板献血员铁蛋白低值率随年龄和献血频次增加呈上升趋势。结论 男性年内多次单采血小板献血人群中低铁蛋白率增高明显,应引起采供血机构和献血员重视。

Serum ferritin and the risk of early-onset colorectal cancer

BACKGROUND The incidence of early-onset colorectal cancer(EO-CRC)is rising in the United States,and is often diagnosed at advanced stages.Low serum ferritin is often incidentally discovered in young adults,however,the indication for endoscopy in EO-CRC is unclear.AIM To compare serum ferritin between patients with EO-CRC and healthy controls(HCs),and examine the association of serum ferritin in EO-CRC with patient-and disease-specific characteristics.METHODS A retrospective study of patients<50 years with newly-diagnosed EO-CRC was conducted from 1/2013-12/2023.Patients were included if serum ferritin was measured within 2 years prior to 1 year following CRC histologic diagnosis.To supplement the analysis,a cohort of HCs meeting similar inclusion and exclusion criteria were identified for comparison.A sensitivity analysis including only patients with serum ferritin obtained at or before diagnosis was separately performed to minimize risk of confounding.RESULTS Among 85 patients identified with EO-CRC(48 females),the median serum ferritin level was 26 ng/mL(range<1-2759 ng/mL).Compared to HCs(n=80211),there were a higher proportion of individuals with EO-CRC with serum ferritin<20 ng/mL(female 65%,male 40%)versus HCs(female 32.1%,male 7.2%)age 29-39 years(P=0.002 and P<0.00001,respectively).Stage IV disease was associated with significantly higher serum ferritin compared to less advanced stages(P<0.001).Serum ferritin obtained before or at the time of diagnosis was lower than levels obtained after diagnosis.Similar findings were confirmed in the sensitivity analysis.CONCLUSION Severe iron deficiency may indicate an increased risk of EO-CRC,particularly at earlier stages.Further studies defining the optimal serum ferritin threshold and routine incorporation of serum ferritin in screening algorithms is essential to develop more effective screening strategies for EO-CRC.

Iron and ferritin effects on intensive care unit mortality:A metaanalysis被引量:1

BACKGROUND The effect of serum iron or ferritin parameters on mortality among critically ill patients is not well characterized.AIM To determine the association between serum iron or ferritin parameters and mortality among critically ill patients.METHODS Web of Science,Embase,PubMed,and Cochrane Library databases were searched for studies on serum iron or ferritin parameters and mortality among critically ill patients.Two reviewers independently assessed,selected,and abstracted data from studies reporting on serum iron or ferritin parameters and mortality among critically ill patients.Data on serum iron or ferritin levels,mortality,and demographics were extracted.RESULTS Nineteen studies comprising 125490 patients were eligible for inclusion.We observed a slight negative effect of serum ferritin on mortality in the United States population[relative risk(RR)1.002;95%CI:1.002-1.004].In patients with sepsis,serum iron had a significant negative effect on mortality(RR=1.567;95%CI:1.208-1.925).CONCLUSION This systematic review presents evidence of a negative correlation between serum iron levels and mortality among patients with sepsis.Furthermore,it reveals a minor yet adverse impact of serum ferritin on mortality among the United States population.

基于ferritin纳米载体的抑制素α亚基的多克隆抗体的制备

为了制备高效价和多动物适用性的抑制素α(inhibinα,INH-α)多克隆抗体,通过生物信息学分析INH-α物种同源保守性,并以pET-32α为骨架,构建INH-α和INH-α-Ferritin表达载体,原核表达纯化后的重组蛋白用于免疫日本大耳兔,采用间接ELISA方法测定兔血清中抗体效价。结果表明,本研究成功构建了INH-α和INH-α-Ferritin原核表达载体,在0.4 mmol/L IPTG、37℃诱导6 h的条件下,分别获得了35 kDa和50 kDa包涵体形式的重组蛋白。通过免疫获得了高效价的兔抗多克隆抗体,INH-α-Ferritin抗血清效价达到1∶19531250。生物信息学分析结果显示INH-α在哺乳动物间的同源性高、保守性强。本研究成功构建INH-α和INH-α-Ferritin原核表达载体,并通过INH-α-Ferritin显著提高多克隆抗体的效价,为提高哺乳动物家畜繁殖力的研究奠定基础。

Ferritin and mortality in hemodialysis patients with COVID-19:A systematic review and meta-analysis

Objective:To investigate the difference in serum ferritin levels between deceased and surviving regular hemodialysis patients with COVID-19.Methods:We conducted a systematic search across four databases following the PRISMA statement guidelines.Studies reporting ferritin levels and mortality of regular hemodialysis patients with COVID-19 were included.Employing the random-effects model,we performed a meta-analysis to determine the mean difference in serum ferritin levels between the studied groups,along with their corresponding 95%confidence intervals.The meta-analysis was carried out using Review Manager 5.4 and Stata 16.Results:A total of 1013 patients from seven studies were included in this study.Our meta-analysis showed higher mean serum ferritin in the deceased compared to surviving regular hemodialysis patients with COVID-19,with a mean difference of 449.43 ng/mL[95%CI(244.07,654.80),P<0.0001;I2=58%,P=0.003].Conclusions:Our study found a higher mean of serum ferritin levels in the deceased compared to surviving regular hemodialysis patients with COVID-19.

AFP、PIVKA-Ⅱ、Ferritin联合应用在肝癌诊断及疗效监测中的效能评价

目的评价单独及联合应用甲胎蛋白(AFP)、异常凝血酶原(PIVKA-Ⅱ)、铁蛋白(Ferritin)指标在肝癌诊断及预后评估中的效能。方法回顾性分析10个月内在苏州大学附属第二医院核医学科就诊的体检、门诊、住院患者的AFP、PIVKA-Ⅱ、Ferritin检测数值,按照临床诊断信息分为健康体检组(74例)、肝炎肝囊肿组(63例)、肝硬化组(40例)、腺瘤增生组(76例)及肝癌组(788例),比较各指标在各肝相关分组中的表达水平差异。通过Passing-Bablok回归性分析及Kappa分析评价AFP与PIVKA-Ⅱ的应用特性。通过ROC曲线及Kappa分析比较各指标单独及联合应用在诊断肝恶性肿瘤的效能。比较各指标对肝恶性肿瘤预后的评估价值。结果3指标在各分组中的表达差异明显,AFP在健康人群阳性检出率为4.05%,在肝癌组阳性检出率为57.61%;PIVKA-Ⅱ在健康体检组阳性检出率为4.05%,在肝癌组阳性检出率为64.47%;Ferritin在健康体检组阳性检出率为12.16%,在肝癌组阳性检出率为34.90%。分析AFP与PIVKA-Ⅱ指标相关性,Y=0.0786 X+1.5675,线性度偏差显著;一致性分析,Kappa值为0.381,一致性一般。多指标联合应用在肝癌组的诊断效能高于单一指标,ROC曲线下面积为0.802,在腺瘤增生+肝癌组中诊断的ROC曲线下面积为0.771;在肝癌诊断中,AFP、Ferritin、PIVKA-Ⅱ3项联检的灵敏度为88.96%,特异性为80.24%,高于单项目检测的灵敏度,在良恶性肿瘤诊断中,3项联检的灵敏度为82.52%,特异性为78.53%。3指标在肝癌治疗前后差异明显,AFP在治疗前后中位数由4.11ng/mL下调至3.2ng/mL,Ferritin在治疗前后中位数由244ng/mL下调至223ng/mL,PIVKA-Ⅱ在治疗前后中位数由161mIU/mL下调至24.78mIU/mL;秩和检验分析治疗前后数值差异,AFP:P<0.0247;Ferritin:P<0.4842;PIVKA-Ⅱ:P<0.0001,AFP、PIVKA-Ⅱ指标变化有更强的临床一致性和预后指导价值。结论3项传统肝癌检测指标在良恶性疾病中表达差异明显,其�

Microneedle patch loaded with ferritin-nanocaged doxorubicin for locally targeted drug delivery and efficient skin cancer treatment

Ferritin has emerged as a promising nanocarrier for delivering therapeutic agents to tumours.However,the limited drug loading and the off-target impacts after systemic administration remain challenges for cancer treatment with ferritin-based agents.Herein,we develop a microneedle patch loaded with ferritin-nanocaged doxorubicin(DoxFe@Fn/MN)for skin cancer treatment.Briefly,doxorubicin(Dox)is encapsulated in ferritin(Fn)using an iron core-assisted strategy,which results in a 3.4-fold increase in Dox loading compared to the direct loading method.Then,a polyvinyl alcohol-based microneedle(MN)patch is used for the transdermal delivery of DoxFe@Fn,enabling targeted tumour accumulation of DoxFe@Fn and preventing off-target impacts.The released DoxFe@Fn can bind to CD71 highly expressed on skin cancer cells,facilitating its uptake.As a result,the DoxFe@Fn/MN therapy presents a robust antitumour effect in a melanoma tumour model,showing its potential as a promising therapeutic modality for skin cancer treatment.

BACKGROUND Gastric cancer is a kind of malignant tumor which is prevalent all over the world.Although some progress has been made in the treatment of gastric cancer,its prognosis is still not optimistic,so it is of great significance to find reliable prog-nostic indicators to guide the treatment and management of patients with gastric cancer.AIM To explore the relationship between serum levels of five biomarkers[carcinoem-bryonic antigen(CEA),carbohydrate antigen(CA)19-9,CA72-4,CA24-2,and ferritin]and prognosis in patients with gastric cancer.METHODS This study included 200 patients with gastric adenocarcinoma,and conducted an in-depth analysis of their baseline characteristics,relationship between tumor markers and staging,and prognosis.The study found that CA19-9 has a signi-ficant correlation with tumor stage,the average levels of CA24-2,CEA,CA72-4 and ferritin were slightly increased disregarding the stage of tumor.Survival analysis showed that increases in CEA,CA19-9,CA24-2,and ferritin were all associated with shortened overall survival of patients.Further multivariate ana-lysis revealed that elevated serum CA72-4 levels were an inde-pendent adverse prognostic factor.RESULTS This study reveals that there is a significant correlation between the expression levels of serum tumor markers CEA,CA19-9,CA72-4,CA24-2 and ferritin in patients with gastric cancer and prognosis,and can be used as important indicators for prognostic evaluation of gastric cancer.In particular,markers that appear abnormally elevated initially may help identify gastric cancer patients with poor prognosis.CONCLUSION Serum CEA and CA19-9 play an important role in the prognosis assessment of gastric cancer,and are effective tools to guide clinical practice and optimize individualized treatment strategies for gastric cancer patients.

线粒体铁蛋白在神经系统疾病的研究进展

线粒体铁蛋白(FtMt)是一种无内含子基因编码的铁蛋白,能通过调控细胞氧化应激、胞质铁的平衡,进而影响线粒体铁含量、线粒体功能、活性氧的产生和细胞GSH含量等。越来越多的证据表明FtMt参与神经退行性病变、缺血性脑卒中、创伤性脑损伤、不安腿综合征以及脑胶质瘤的发生、发展,本文总结了FtMt在多种神经系统疾病中发挥的不同作用,为探索多种神经系统疾病的机制研究提供参考。

铁蛋白纳米颗粒体内成像及肿瘤靶向性研究

目的:构建近红外荧光探针修饰的铁蛋白纳米颗粒,研究其作为体内成像系统的安全性以及在小鼠体内的生物分布情况和对肿瘤组织的靶向性。方法:通过在铁蛋白(ferritin)表面人工设计半胱氨酸点突变,通过半胱氨酸巯基和含有马来酰亚胺(maleimide)的Dy731近红外荧光探针分子进行巯基偶联,对铁蛋白纳米颗粒表面进行功能化修饰,使荧光探针以共价的形式结合在蛋白纳米颗粒的表面;将近红外荧光探针修饰过的铁蛋白纳米颗粒(Dy731-Ft)与5%w/v琼脂糖溶液混合冷却后制成凝胶,置于小鼠皮下和腹腔肌肉壁下,检测蛋白纳米颗粒穿透皮肤和肌肉组织的能力;将Dy731-Ft蛋白纳米颗粒通过尾静脉对小鼠进行活体注射,观察小鼠是否产生急性不良反应,检测近红外荧光强度以观察Dy731-Ft作为体内成像系统在小鼠体内的分布情况;构建U87-MG肿瘤异种移植动物模型,进行尾静脉注射验证Dy731-Ft在荷瘤小鼠组织器官内的分布和对于肿瘤组织的靶向性。结果:通过动物实验表明,小鼠体内注射铁蛋白纳米颗粒和显像期间及之后均未见急性不良反应,说明铁蛋白纳米颗粒具有良好的生物相容性,进入体内主要在肝脏积累。结论:近红外Dy731-Ft蛋白纳米颗粒能够靶向U87-MG肿瘤组织,推测其在肿瘤组织内的积累可能与转铁蛋白受体1(transferrin receptor 1,TfR-1)介导的细胞内吞作用和肿瘤组织增强渗透性以及滞留效应(enhanced permeability and retention effect,EPR)的协同作用相关。
